5-HT3-Antagonist

5-HT3-Antagonisten („Setrone“) sind Arzneistoffe, die selektiv 5-HT3-Rezeptor (Serotonin-Rezeptor) als Hemmstoffe besetzen. Sie werden als Antiemetika zur Behandlung von durch Zytostatika oder Bestrahlung ausgelöster Übelkeit und Erbrechen eingesetzt. Durch den Einsatz der 5-HT3-Antagonisten wird die Lebensqualität von Patienten während einer Tumor-Chemotherapie oder Strahlentherapie erheblich verbessert. Des Weiteren kommen sie zur Therapie von postoperativer Übelkeit und Erbrechen zum Einsatz.

Eine Ausnahme in dieser Wirkstoffklasse bildet Alosetron, das in den USA unter dem Handelsnamen Lotronex zur Behandlung des Reizdarmsyndroms zugelassen ist.[1] Für den Vertreter Tropisetron gibt es durch mehrere Studien Hinweise für eine Verminderung der Schmerzen bei dem bisher schwer behandelbaren Fibromyalgiesyndrom.[2] Dies gilt allerdings nur für einen Teil der Patienten, da andere Patienten für das Medikament als therapieresistent eingestuft werden.[3]
