Ligase

Ligasen (v. lat. ligare "verbinden", "verketten") sind Enzyme, die das Verknüpfen zweier Moleküle durch eine chemische Bindung katalysieren. Dazu benötigen sie Energie, die aus der Spaltung energiereicher Nukleosidtriphosphate (NTP) stammt. Dabei werden ein oder zwei Phosphatreste abgespalten. Auch andere Moleküle wie NAD+ können als Energielieferant dienen. Da diese jedoch unter anderem durch ATP-Verbrauch regeneriert werden können, gelten die dadurch mit Energie belieferten Enzyme ebenfalls als "Ligasen".

Schematisch verläuft die Ligase-Reaktion folgendermaßen (M1, M2 sind die zu verbindenden Moleküle) ab:

Klassifikation

Ligasen werden in der EC-Nummern-Klassifikation mit der Nummer 6 bezeichnet. Ein spezielles Ligase-Enzym hat demnach die Nummer 6.X.X.X. Das zweite X definiert die genauere Art der Verknüpfung:

  • EC 6.1 Verknüpfung von Kohlenstoff und Sauerstoff
  • EC 6.2 Verknüpfung von Kohlenstoff und Schwefel
  • EC 6.3 Verknüpfung von Kohlenstoff und Stickstoff
  • EC 6.4 Verknüpfung zweier Kohlenstoffatome
  • EC 6.5 Bildung eines Phosphoresters
  • EC 6.6 Bildung einer Stickstoff-Metallatom-Bindung

Falsche Freunde und bedeutende Gruppen

Der Begriff Ligase wird häufig synonym für eine spezielle Untergruppe, die DNA-Ligasen verwendet, was aber nicht ganz korrekt ist. DNA-Ligasen verknüpfen (ligieren) zwei DNA-Stränge, indem eine Phosphor-Diesterbindung hergestellt wird. Somit gehören DNA-Ligasen in die Klasse 6.5.

Der Begriff Synthetase wird ebenfalls häufig synonym gebraucht, ist aber ebenso falsch, da Synthetasen nur ATP und nicht allgemein NTPs verwenden. Synthetasen sollten auch nicht mit Synthasen verwechselt werden, die keine zusätzliche Energie für die Additionsreaktion benötigen.

Eine weitere in der Biochemie bedeutende Gruppe sind die Carboxylasen. Diese Enzyme lagern Kohlenstoffdioxid in Form von Karbonationen an andere Moleküle (EC 6.4.1.).
