Die Gattung Orthopoxvirus besitzt eine lineare doppelsträngige DNA (130 bis 375Kb) und hat eine Größe von 170 bis 450 nm. Orthopoxvirusinfektionen werden in der Tierseuchengesetzgebung auch als Säugerpocken bezeichnet.

Sie umfasst folgende Arten mit deren Varietäten:

• Orthopoxvirus variola = Variolavirus (Endwirt Mensch), → Pocken
  • Orthopoxvirus variola var. alastrim = Kaffernpockenvirus (Endwirt Mensch)
• Orthopoxvirus vaccinia = Vacciniavirus (Endwirte sämtliche Säugetiere), → Pocken
  • Orthopoxvirus vaccinia var. equi = Pferdepockenvirus (Endwirte Pferd, Esel)
  • Orthopoxvirus vaccinia var. bubali = Büffelpockenvirus (Endwirt Büffel)
  • Orthopoxvirus vaccinia var. cuniculi = Kaninchenpockenvirus (Endwirt Kaninchen)
• Orthopoxvirus bovis = Kuhpockenvirus (Endwirte Rind, Schaf, Ziege, Katze, Hund, wildlebende Nagetiere), → Kuhpocken
  • Orthopoxvirus bovis var. elefanti (Endwirt Elefant)
• Orthopoxvirus muris = Ektromelievirus (Endwirte Maus, Fuchs, Nerz)
• Orthopoxvirus cameli = Kamelpockenvirus (Endwirt Kamel)
• Orthopoxvirus simiae = Affenpockenvirus (Endwirt Affe), → Affenpocken
