Turnover Frequency

Die Wechselzahl (engl. Turn over number, abk. TON) beschreibt in der Chemie die Anzahl an Formelumsätzen, die ein bestimmter Katalysator beschleunigen kann. Sie ist ein Maß für die Effizienz eines Katalysators und bestimmt das größtmögliche stöchiometrische Verhältnis zwischen Edukt und Katalysator, bei der das Edukt noch vollständig umgesetzt wird, bevor der Katalysator selbst inaktiv wird.

Abgrenzung

In der Biochemie charakterisiert die Wechselzahl, zusammen mit der Michaelis-Menten-Konstante die Leistungsfähigkeit eines einzelnen Enzymmoleküls. Bei enzymatischen Reaktionen wird anstelle der Gesamtzahl an möglichen Umsätzen die Anzahl der Umsätze pro Zeiteinheit bestimmt. Damit ist die Wechselzahl in diesem Zusammenhang die Geschwindigkeitskonstante der enzymatischen Reaktion bei einer Enzymkonzentration von 1 mol/l. Daher wird hier auch von molekularer Aktivität (kcat) gesprochen. Mathematisch betrachtet beschreibt der Wert also das Maximum des Enzymumsatzes pro Zeit, der durch weitere Substratgabe nicht mehr steigerbar ist.

Beispiele aus der Biochemie

Man erhält die Wechselzahl (kcat) durch Bestimmung der Aktivität (Vmax) dividiert durch die Enzymkonzentration (mol/l) im Test. Sie hat die Dimension k-1 (s-1). Wechselzahlen bewegen sich im Bereich von etwa 0,5 (Lysozym) und 1.000.000 (Carboanhydrase). Der Quotient aus Wechselzahl (kcat) und Michaelis-Menten-Konstante (Km) beschreibt die katalytische Effizienz.
